All files and folder in folder %windir% and the directory itself are read only, but it doesn't mean, that there isn't any programs or users, who cannot make changes or delete it at all. I'd suggest using some other method/s to block apps. & programs from calling home, instead of fiddling with the hosts file — remember . . . . . it lacks the capability to block IPs. Personally, I enjoy a finer granular control by using the following to block hosts, domains and IPs — they are also tamper-free and block at a very early stage:— Acrylic Proxy (hosts + domains,) Simple DnsCrypt (hosts + domains,) AdGuard (hosts + domains + IPs,) and Windows 10 Firewall Control (hosts + domains + IPs.)
