If your booting on a USB 2.0 cable with a slow hd (say 5200 rpm) then yes boot up will be slower, booting with a USB3.0 7200rpm disk will still be slower than plugging directly into the motherboard. I have 3 x 2 tb externals and my startup is slow... <have to be external. so internal space. well over the 1 min range.. about 73seconds.. i also have 2 x 3tb and 2 1.5tb, 1 x 1tb and a dvd burner internals.. i only have 6 sata connections on my motherboard all used..
